You should be able to drive again as normal on a proper plug, but getting the correct seal and ensuring a plug is the correct repair for the issue is crucial. However, a plug is still intended to be a temporary measure and it may only last until the next encounter with road debris or major change in air pressure. A tire plug may be the right solution for your tire issue if you have a puncture in the tread, NOT the sidewall, that is less than ¼” in diameter. (For reference, a plain M&M candy is about double the maximum size.) For larger punctures, a patch may be appropriate; depending on the severity or if the damage is on the sidewall or certain interior layers of the tire, replacing it may be the only safe option to prevent a potential blowout. 

A tire plug is intended to be a temporary measure, and a patch tends to offer a longer term solution. While some tire manufacturers claim that a properly patched tire (with small tread puncture) can last many thousands of miles, that’s not the case if there isn’t a proper seal or if the repair is performed on a puncture that’s too large or deep. If you find that you keep encountering punctures on the same tire, which may necessitate replacement, an alignment service and ensuring that both tires along an axle are wearing evenly could prevent further damage.
